Polynucleotide kinase (PNK) is an enzyme that catalyzes the reversible phosphorylation of nucleoside monophosphates, ss and ds nucleic acids. PNK from bacteriophage T4 is widely used in molecular biology for 5′ phosphorylation of DNA and RNA for the purpose of labeling and ligation, as well as the removal of 3′phosphoryl groups from various nucleotide and nucleic acid substrates for labeling purposes.
